THÈMES DE RECHERCHE

Mes activités de recherche portent sur l'estimation de la diversité moléculaire et génomique pour les fins de l'analyse phylogénétique, la biosystématique, la biogéographie et la génétique des populations chez les espèces végétales et les essences forestières. La cartographie du génome des conifères et la sélection assistée par marqueurs font également partie de mes préoccupations. Des méthodes d'analyse numérique de la diversité génétique et de nouveaux types de marqueurs moléculaires sont aussi développées.

RÉALISATIONS RÉCENTES

Écologie moléculaire

Nos récents efforts en écologie moléculaire ont porté sur le développement d'une nouvelle génération de marqueurs moléculaires de régions codantes du génome, les ESTPs (Expressed Sequence Tag Polymorphisms).

Les marqueurs que nous avons développés sont actuellement utilisés par plusieurs équipes en Europe et aux États-Unis. En plus de mettre au point ces marqueurs pour un nombre important d'espèces conifériennes, nous avons pu les utiliser à diverses fins en écologie moléculaire. Nous avons ainsi pu démontrer, par analyse génomique, que la diversité génétique de l'épinette rouge (Picea rubens) était fortement réduite par rapport à sa proche cousine, l'épinette noire (Picea mariana), en formant un sous-ensemble parfait. L'analyse numérique des données a permis de démontrer que l'épinette rouge aurait divergé de l'épinette noire aussi tard que vers la fin de l'ère du Pléistocène, lors d'une des dernières avancées glaciaires, faisant de l'épinette rouge probablement la plus jeune espèce coniférienne de l'Est de l'Amérique du Nord. Cette origine récente pourrait expliquer sa faible amplitude écologique et sa distribution naturelle restreinte.

Ces mêmes marqueurs ont également permis de démontrer que la coupe avec protection de la régénération préétablie de marcottes d'épinette noire ne résultait pas en des peuplements au niveau augmenté de consanguinité par rapport aux peuplements régénérés naturellement par semences après feu, et ce, après un cycle de récolte. De même, les niveaux de croisements consanguins (endogamie) n'étaient pas augmentés. La CPRS ne semble donc pas entraîner d'effets dysgéniques pour la période étudiée. D'autre part, nous avons pu démontrer, avec nos collègues du Centre d'études nordiques, que certaines épinettes noires prostrées de la toundra s'étaient maintenues pendant une période de près de 2 000 ans. C'est par marcottage successif, i.e. par clonage naturel, que ces individus ont réussi à atteindre ce vénérable âge record. D'autre part, la diversité génétique de ces populations fossiles n'était pas réduite, par rapport aux populations contemporaines issues de feux que l'on retrouve plus au sud.

Sur un autre front, nous avons également pu démontrer l'incidence élevée de l'hybridation naturelle au sein des peuplements mixtes d'épinette rouge et d'épinette noire de la vallée du St-Laurent, possiblement en raison du fort niveau de perturbation du paysage naturel dans cette région. La reconnaissance des espèces pures et des hybrides qui s'appuyait uniquement à partir de l'analyse de caractères morphologiques n'était pas fiable. Il en résulte que l'on devrait s'abstenir de récolter les semences dans ces peuplements mixtes.

D'autres études chez le noyer cendré (Juglans cinerea) et chez l'if du Canada (Taxus canadensis), en collaboration avec le Service canadien des forêts (SCF), ont permis de démontrer les niveaux exceptionnellement bas de diversité génétique chez ces espèces. Une relation avec la faible abondance de ces espèces dans l'écosystème est probable, suggérant que la survie de ces espèces est bien plus au niveau de leur maintien au niveau du paysage (métapopulation) qu'au niveau local. Les implications sont importantes pour les stratégies de conservation.

Génomique évolutive et structurale

Nos études du génome mitochondrial chez les arbres et les plantes annuelles ont démontré l'évolution plus lente chez les espèces pérennes comme les arbres, confirmant une tendance que nous avions identifiée précédemment à partir de l'analyse du génome chloroplastique. Nous avons également dressé la carte phylogéographique mondiale du genre Fraxinus à l'aide de séquences d'ITS ribosomique, et identifié plusieurs migrations entre l'Amérique du Nord, centre d'origine du genre, et l'Asie. Ces migrations ont été suivies d'une dispersion du genre de l'Asie vers l'Europe avec des spéciations successives. Chez les conifères, nous avons dressé les premières cartes du génome nucléaire de l'épinette blanche (Picea glauca), et sommes en voie de compléter ce travail chez l'épinette noire.

Les travaux effectués avec nos collaborateurs du SCF dans le cadre du Consortium international sur le génome des conifères ont permis de placer près de 200 marqueurs sur chacune de deux cartes chez l'épinette blanche. Ces cartes permettront d'obtenir une carte consensus pour chaque espèce ainsi que pour le genre Picea. Pour l'heure, les cartes chez l'épinette blanche démontrent une longueur supérieure à celle observée par nos collègues américains et français pour les cartes d'espèces de pin.
