Résumé
In this study, Agrobacterium -mediated transformations of black spruce ( Picea mariana ) and hybrid poplar ( Populus nigra × Populus maximowiczii ) were accomplished in attempt to increase their resistance against fungal pathogens. Both species were transformed with the endochitinase gene ( ech42 ) from the biocontrol fungus Trichoderma harzianum . Fifteen transgenic black spruce and six poplar lines were obtained covering a range in the levels of ech42 expression. In vitro pathogenesis tests were conducted using foliar discs from transgenic poplars. These transgenic poplars showed increased resistance to the fungal pathogen Melampsora medusae while untransformed poplars were normally infected. The transgenic spruce seedlings also showed increased resistance to the spruce pathogen Cylindrocladium floridanum . These results show that constitutive expression of the ech42 gene from Trichoderma harzianum could be exploited to enhance resistance to fungal pathogens in forest tree species.
